Product Specific Information

  • Reconstitution: Reconstitute with 0.5 mL of distilled water.
  • Dilution: Use a buffer containing a carrier protein (e.g., PBS with 1% BSA) or a neutral medium containing 10% FCS. Freeze aliquots at -70°C.
  • Activity: The biological activity was measured using L929 mouse fibrosarcoma cells with an ED50 of 5–8 ng/mL.
  • Expression: The mature, soluble form of bovine TNF-alpha was expressed in Pichia pastoris KM71H and therefore does not contain endotoxin.

Target Information

TNF alpha is a multifunctional proinflammatory cytokine belonging to the tumor necrosis factor (TNF) superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cells, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.

It exists as multimers (dimers, trimers, or pentamers) but shows a single 17 kDa band under non-reducing SDS-PAGE conditions. Knockout studies in mice suggest a neuroprotective function, and TNF alpha causes tumor necrosis when injected into tumor-bearing mice. It also plays roles in immune responses to bacterial, viral, parasitic, and fungal infections, as well as in tumor cytolysis or cytostasis, often synergistic with interferon-gamma.

This cytokine has been implicated in autoimmune diseases, insulin resistance, and cancer.
