Target Information
Tuberous sclerosis complex (TSC) is an autosomal dominant tumor syndrome caused by mutations in either the TSC1 or TSC2 tumor suppressor genes. The products of these genes form a protein complex that indirectly decreases the signaling of mammalian Target of Rapamycin (TOR), a serine/threonine kinase regulating cell growth and cycle through nutrient and growth factor signals.
TOR activity is stimulated by Rheb, a Ras superfamily G-protein, when the GTP/GDP ratio bound to Rheb is high. Immunoprecipitated TSC1/TSC2 complex stimulates Rheb GTPase activity in vitro, reducing Rheb’s ability to activate TOR. Overexpression of TSC1 and TSC2 decreases the GTP/GDP ratio bound to Rheb and inhibits cell growth. A shorter 40 kDa isoform of TSC1 also exists, though its function remains unknown.
