Target Information

Oncogene ERBB4 (HER4) is a member of the type I receptor protein tyrosine kinase subfamily that includes EGFR, ERBB2, and ERBB3. ERBB4 binds and is activated by neuregulins and other factors, inducing cellular responses such as mitogenesis and differentiation.
ErbB4 acts as a receptor for heregulin and mediates HGL-stimulated tyrosine phosphorylation, regulating key processes including cellular metabolism, transcription, cell cycle progression, cytoskeletal rearrangement, cell movement, apoptosis, and differentiation.

Expression of ErbB4 is most predominant in several breast carcinoma cell lines and normal tissues such as skeletal muscle, heart, pituitary, brain, and cerebellum. Breast tumor cell lines T47-D, MDA-MB-453, BT-474, and H3396 show high mRNA levels, while MCF-7, MDA-MB-330, and MDA-MB-361 show intermediate levels. Low or absent expression is observed in MDA-MB-231, MDA-MB-157, MDA-MB-468, and SKBR-3. Elevated ErbB4 levels in certain tumor lines suggest a possible role in malignancy.
