Target Information
DNA-directed RNA polymerase II subunit RPB1 (POLR1A)는 DNA 의존성 RNA 중합효소로, 네 가지 리보뉴클레오타이드 삼인산을 기질로 사용하여 DNA를 RNA로 전사합니다. POLR1A는 RNA polymerase II의 가장 큰 소단위이며, mRNA 전구체 및 다양한 비암호화 RNA를 합성하는 촉매 성분입니다.
이 단백질은 RNA polymerase II 전사 기계의 중심 구성요소로, 상호 이동하는 여러 구조적 요소로 이루어져 있습니다. RPB1은 DNA 템플릿을 잡는 핵심 요소로, 전사 개시 시 프로모터의 단일가닥 DNA가 활성 부위 틈에 위치하게 합니다.
전사 신장 동안 Pol II는 템플릿을 따라 이동하며, C-말단 도메인(CTD)의 인산화 상태에 따라 전사 개시, 신장, 종결, mRNA 가공 조절 인자들의 결합이 조절됩니다. 유전자 발현 수준의 조절은 CTD 라이신의 메틸화 및 아세틸화 수준의 균형에 따라 결정됩니다.
