Target Information
MAP4K1 (also known as HPK1, Hematopoietic Progenitor Kinase 1) is a hematopoietic cell-restricted member of the Ste20 serine/threonine kinase superfamily. It acts as a tissue-specific upstream activator of the MEKK/JNK/SAPK signaling pathway.
HPK1 reduces T cell receptor (TCR) signaling and T cell proliferation by phosphorylating the adaptor protein SLP-76. JNKs/SAPKs play a crucial role in stress responses in mammalian cells.
HPK1 is expressed predominantly in hematopoietic cells, including early progenitor cells, and specifically activates JNK1, enhancing AP-1-mediated transcriptional activity. It binds and phosphorylates MEKK1 directly, but does not interact with Rac1 or Cdc42, distinguishing it from PAK65.
