Target Information
The second messengers cAMP and cGMP are key regulatory molecules involved in diverse signal transduction pathways. Their levels are controlled by synthesis (nucleotide cyclases) and hydrolysis (cyclic nucleotide phosphodiesterases, PDEs).
PDEs catalyze the conversion of cyclic nucleotides to nucleoside monophosphates. PDE6 acts as the effector enzyme in the G protein-mediated visual signal transduction cascade.
PDE6 consists of multiple subunits: rod and cone-specific catalytic subunits (alpha’, alpha, beta), inhibitory subunit gamma, and delta of unknown function. The catalytic core forms alpha’/alpha’ homodimers in cones and alpha/beta heterodimers in rods.
C-terminal modifications such as methylation, myristoylation, and prenylation are essential for complex assembly and membrane association.
