Background

The 26S proteasome is a multicatalytic proteinase complex with a highly ordered structure composed of two complexes: a 20S core and a 19S regulator.

  • The 20S core consists of four rings of 28 non-identical subunits (two rings of 7 alpha subunits and two rings of 7 beta subunits).
  • The 19S regulator comprises a base (6 ATPase subunits and 2 non-ATPase subunits) and a lid (up to 10 non-ATPase subunits).

Proteasomes are abundant in eukaryotic cells and cleave peptides in an ATP/ubiquitin-dependent, non-lysosomal pathway.
A modified form, the immunoproteasome, plays an essential role in processing class I MHC peptides.

This gene encodes the largest non-ATPase subunit of the 19S regulator lid, responsible for substrate recognition and binding. Alternatively spliced transcript variants have been identified for this gene.
