Product Specific Information

The ISA-3 monoclonal antibody reacts with human ICOS (Inducible COStimulatory molecule), also known as H4, CRP-1, and AILIM. ICOS is a T cell activation molecule and a member of the CD28/CTLA-4 family. It plays a critical role in humoral immune responses and T cell activation.

Plate-bound ISA-3 induces production of IL-4, IL-5, IL-10, and other cytokines but not IL-2. It was generated against human ICOS antigen.
C398.4A (anti-mouse ICOS/H4) cross-reacts with human ICOS but binds a different epitope. For brightest staining of activated human T cells, use Product #13-9948-82 or #12-9948-42.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on unstimulated and CD3/CD28-stimulated human blood cells.
Use ≤ 5 µL (0.6 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test). Antibody should be titrated for optimal performance.

Note:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Product #C001T06F01):
Required for optimal staining with NovaFluor conjugates. Add 5 µL per sample to antibody cocktails. For single-color controls, use 5 µL per 100 µL cell sample.

NovaFluor conjugates utilize Phiton™ technology based on novel nucleic acid dye structures for engineered fluorescent signatures minimizing spillover and spread.
