Background
The secreted polypeptide encoded by this gene binds and inactivates members of the transforming growth factor-beta (TGF-β) superfamily signaling proteins, such as bone morphogenetic protein-4 (BMP4). By diffusing through extracellular matrices more efficiently than members of the TGF-β superfamily, this protein may play a principal role in creating morphogenic gradients. It has pleiotropic effects both early in development and in later stages. Originally isolated from Xenopus for its ability to restore normal dorsal-ventral body axis in UV-ventralized embryos, knockout studies in mice suggest involvement in neural tube fusion and joint formation. Dominant human NOG mutations have been linked to proximal symphalangism (SYM1) and related disorders.
