Target Information

EIF2AK2 (PKR) is one of four kinases that specifically phosphorylate Ser51 of translation initiation factor eIF2-alpha in response to various environmental stresses. EIF2AK2 plays a key role in antiviral defense mediated through its direct activation by double-stranded RNA produced by viral infection. PKR is critical in IFN-induced innate antiviral response, virus-induced apoptosis, and cell growth & differentiation. It induces apoptosis by up-regulating Fas expression and mediates the FADD/Caspase-8 death signaling pathway.

Upon viral infection, PKR functions as a dual protein, sequentially activating both cell survival and cell death pathways using kinase-independent and dependent strategies. PKR regulates multiple signaling pathways, including NF-kappaB activation, p53, p38, and PDGF pathways. It has been implicated in tumor suppression and malignancy. Viruses have evolved mechanisms to evade PKR’s antiviral effects, such as inhibition by influenza virus NS1 protein. PKR is also associated with several neurodegenerative diseases, including Alzheimer’s, Huntington’s, and amyotrophic lateral sclerosis.
