Product Specific Information

This AD2 monoclonal antibody reacts with human CD73, a 5'-ectonucleotidase that converts 5'-adenosine monophosphate to adenosine. CD73 is expressed on endothelial cells, B cells, and T cells, including some CD4+Foxp3+ regulatory T cells. Adenosine production by these cells inhibits CD4 T cell effector functions such as proliferation and cytokine secretion.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells.
Use 5 µL (0.4 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability detection.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample to antibody cocktails before combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample containing 10³–10⁸ cells.

Target Information

CD73 (Ecto-5'-nucleotidase) catalyzes the conversion of purine 5'-mononucleotides to nucleosides, preferring AMP as a substrate. It forms a dimer of two 70 kDa subunits attached to the plasma membrane via a glycosyl phosphatidyl inositol linkage. CD73 serves as a lymphocyte differentiation marker, and its deficiency is linked to various immunodeficiency diseases. The CD73 gene is localized to chromosome 6q14–q21. Defects can lead to joint and artery calcification and intestinal tuberculosis. Two transcript variants encoding different isoforms are known.
