Product Specific Information

The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule, a 32–34 kDa cell surface receptor expressed as either CD8αβ heterodimer or CD8αα homodimer.

• CD8αβ: expressed on most thymocytes and mature αβ TCR T cells
• CD8αα: expressed on γδ TCR T cells, intestinal intraepithelial lymphocytes (IELs), and dendritic cells

CD8 binds to MHC class I and associates with protein tyrosine kinase p56lck, playing a role in T cell development and activation.

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometry of mouse splenocytes at ≤0.125 µg/test (100 µL final volume). Optimal titration recommended.

Note: Alexa Fluor 561 non-specifically stains B cells in Swiss Webster and SJL mice, but not in BALB/c or C57BL/6 strains.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin family involved in T cell signal transduction, differentiation, and activation.

• Predominantly expressed on T cells as a disulfide-linked heterodimer (CD8αβ) acting as a co-receptor for MHC class I.
• Also expressed as CD8αα homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells.
• Engagement with MHC-I/peptide complexes recruits Lck kinase, promoting CTL activation and immune response.
• Differentiation of naive CD8+ T cells into cytotoxic T lymphocytes is enhanced by IL-2, IL-12, and TGF-β1.
