Immunohistochemistry: Antiserum was used on perfusion-fixed tissue. Perfusion sequence:

1. Calcium-free Tyrode’s solution
2. Paraformaldehyde-picric acid fixative
3. 10% sucrose in PBS (cryo-protectant)

Desired tissues were dissected and stored overnight in 10% sucrose in PBS.
Slide-mounted sections were processed for indirect immunofluorescence:

• Blocking buffer incubation for 1 hour at room temperature
• Primary antiserum diluted in blocking buffer to working concentration
• Incubation for 18–24 hours at 4°C
• Rinse 3 times, then incubate with secondary antibodies for 1 hour at room temperature
• Rinse 3 times, coverslip, and examine under fluorescence microscopy

Note: Sodium azide (NaN₃) interferes with peroxidase reactions and should not be used with peroxidase methodologies. If sodium azide is present, thoroughly rinse tissues with sodium azide-free buffer before peroxidase reaction.

Target Information

Neuropeptide Y (NPY) receptors are G protein-coupled receptors mediating physiological responses such as feeding and vasoconstriction. Endogenous ligands include NPY, peptide YY (PYY), and pancreatic polypeptide (PP). Increased activity of NPY and its receptors (Y1R, Y2R, Y5R) has been observed in experimental obesity. Association studies support a link between NPY2R and obesity.
