Target Information
Glycogen synthase kinase-3 (GSK3) is a protein kinase originally identified as a regulator of glycogen synthase, a key enzyme in glycogen metabolism. It is involved in various cellular functions including protein synthesis, cell proliferation, differentiation, microtubule dynamics, and apoptosis.
GSK3 phosphorylates substrates only if a phosphoserine or phosphotyrosine is present four residues C-terminal to the phosphorylation site. Two isoforms exist, GSK3 alpha and GSK3 beta, which are regulated by phosphorylation. Phosphorylation of GSK3 beta on Ser9 (Ser21 in GSK3 alpha) by protein kinase B (PKB) causes inactivation, while activation requires phosphorylation of Tyr216 (Tyr279 in GSK3 alpha).
Activated GSK3 beta phosphorylates multiple cellular proteins such as p53, c-Myc, c-Jun, HSF-1, and cyclin D1, and contributes to phosphorylation of tau protein, implicated in Alzheimer’s disease progression.
