Target Name: TNK2 (ACK1)
Activated Cdc42-associated kinase (ACK1) is a non-receptor tyrosine kinase that acts downstream of Cdc42. It participates in tumorigenesis and metastasis, particularly in prostate cancer. TNK2 mediates CDC42-dependent cell migration via phosphorylation of BCAR1 and may be involved in synaptic function, plasticity, and brain development. It activates AKT1 (Tyr-176), phosphorylates AR (Tyr-267, Tyr-363), WWOX (Tyr-287), and MCF2, influencing cell signaling and tumor progression. TNK2 regulates AXL receptor levels and contributes to metastatic potential by modulating tumor suppressors (WWOX) and pro-survival factors (AKT1, AR).
