This product is diluted and ready-to-use.
A recommended positive control tissue is tonsil, though other tissues may also be suitable.
The antibody is for professional laboratory use in detecting the corresponding protein in formalin-fixed, paraffin-embedded tissue stained via manual qualitative immunohistochemistry (IHC). It is used after tumor diagnosis by conventional histopathology.
Constant regions (C regions) of immunoglobulins are present in both heavy and light chains and contain carbohydrate attachment sites. They help stabilize variable regions via disulfide bonds and facilitate antigen interaction.
Low IgG4 concentrations are often found in patients with recurrent respiratory tract infections. IgG4-related sclerosing disease is characterized by elevated serum IgG4, fibrosis, and IgG4-positive plasma cell infiltration. IgG4 is overexpressed in inflammatory pseudotumor (IPT) and underexpressed in inflammatory myofibroblastic tumor (IMT). Pulmonary nodular lymphoid hyperplasia (PNLH) shows increased IgG4+ plasma cells.
For IHC, pretreatment with heat-induced or enzymatic epitope retrieval is recommended.
IHC staining visualizes antigens through sequential antibody application, enzyme complex, and chromogenic substrate reactions. Results are evaluated microscopically to aid differential diagnosis.
A positive tissue control must be run with each staining procedure. This control should contain both positive and negative components. External positive controls should be freshly fixed and embedded tissue specimens processed identically to patient samples. Weakly positive tissues are ideal for detecting reagent degradation or sensitivity changes.
Negative control tissues, internal or external, should confirm absence of specific staining and assess non-specific background. If staining occurs in negative controls, patient results are invalid.
Target Information
