Product Specific Information

This D8SEE monoclonal antibody reacts with human CD194 (CCR4). CD194, also known as C-C Chemokine Receptor 4 (CCR4), is a member of the G protein-coupled receptor family characterized by seven transmembrane alpha-helical domains.
Ligands for CCR4 include CCL17 (TARC) and CCL22 (MDC), which direct migration of target cells. CCR4, CCR10, and CLA are important for T cell recruitment into skin and may play roles in adhesion and extravasation.

CCR4 expression is found in Th2, Th17, Treg, and Th1 subsets, but not in naive T cells, indicating its role in memory or effector function. Other CCR4-expressing cells include activated NK cells, platelets, monocytes, and basophils.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry of normal human peripheral blood cells. Use at 5 µL (0.125 µg) per test (100 µL final volume).
Cell number may range from 10⁵ to 10⁸ cells/test.
Excitation: 633–647 nm; Emission: 660 nm; Laser: Red Laser
Filtration: 0.2 µm post-manufacturing filtered.


Target Information

CCR4 is a chemokine receptor preferentially expressed on Th2-type cells. Its expression is transiently increased after TCR and CD28 engagement. Activated Th1 cells also up-regulate CCR4 expression and responsiveness to thymus- and activation-regulated chemokine.

Chemokines are small (8–14 kD), mostly basic molecules regulating leukocyte trafficking via 7-transmembrane, G protein-coupled receptors. They play essential roles in immune system development, homeostasis, and function, and also affect CNS and endothelial cells involved in angiogenesis or angiostasis.

Chemokines are divided into two major subfamilies based on cysteine arrangement:

  • CXC: Two cysteines separated by one amino acid
  • CC: Two adjacent cysteines
