Target Information
The second messengers cAMP and cGMP are key regulatory molecules involved in numerous signal transduction pathways. Their levels are controlled by synthesis via nucleotide cyclases and hydrolysis via cyclic nucleotide phosphodiesterases (PDEs).
PDEs form a superfamily of enzymes that catalyze the conversion of 3′,5′-cyclic nucleotides to nucleoside 5′-monophosphates. PDE6 acts as the effector enzyme in the G protein-mediated visual signal transduction cascade.
PDE6 comprises five subunits:
- Rod and cone-specific catalytic subunits (α′ for cone, α and β for rod)
- Inhibitory subunit γ
- Delta subunit (function unknown; interacts with other proteins)
The catalytic core consists of α′/α′ homodimers (cone) and α/β heterodimers (rod). Both catalytic and inhibitory subunits have C-terminus modifications (methylation, myristoylation, prenylation) essential for complex assembly and membrane association.
