Target Information
Members of the Id family of basic helix-loop-helix (bHLH) proteins include ID1, ID2, ID3, and ID4. These proteins are ubiquitously expressed and dimerize with class A and B HLH proteins. Due to the absence of the basic region, the resulting heterodimers cannot bind DNA, thus negatively regulating DNA binding of bHLH proteins.
ID proteins function to inhibit muscle-specific gene expression by preventing DNA binding of E12 and Myo D. During muscle cell differentiation, reduced ID protein levels allow E12 and/or E47 to interact with Myo D and myogenin, activating myogenic differentiation. Expression of ID proteins is strongly dependent on growth factor activation, and reduction of ID mRNA delays reentry into the cell cycle after growth factor stimulation.
