The RNaseAlert™ QC System v2 detects RNase activity using a convenient and sensitive fluorimetric assay that delivers results in real time. This version features a substrate approximately 60% more sensitive than the previous version and provides reagents sufficient for 5 × 96 (480) high-throughput assays.

작동 원리

This system utilizes a novel RNA substrate tagged with a fluorescent reporter molecule (fluor) on one end and a quencher on the other.

• Without RNase: The quencher suppresses fluorescence to low levels.
• With RNase present: The substrate is cleaved, separating the fluor and quencher, producing a bright green fluorescent signal.

Fluorescence can be detected using filter-based or monochromator-based fluorometers. The substrate sequence is optimized to detect various RNases, including RNase A, RNase T1, RNase I, micrococcal nuclease, S1 nuclease, mung bean nuclease, and Benzonase™.

신속하고 간편한 프로토콜

• Reconstitute the lyophilized fluorescent substrate in the supplied tube.
• Incubate with the test solution for several minutes.
• Measure fluorescence using a fluorometer.
  Samples showing strong fluorescence compared to the negative control indicate RNase contamination. Nuclease-free water and RNase A are included for use as controls.
