Product Specific Information

The monoclonal antibody 51.1 reacts with human CD1d, a member of the CD1 family similar to non-polymorphic MHC Class I-like molecules. CD1d is a conserved single transmembrane receptor of the Immunoglobulin Superfamily. It can associate with beta-microglobulin or exist independently, suggesting different mechanisms for presenting glycolipid molecules to CD1d-reactive NKT cells.

CD1d expression is found on peripheral B cells, resting monocytes, and cortical thymocytes, with polarized expression on intestinal epithelial cells and low intracellular expression in hepatocytes. In HCV-infected livers, CD1d expression is elevated compared to normal controls.

The 51.1 monoclonal antibody has functional activity, blocking the interaction between CD1d-transfected cells and NKT cells.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometry on normal human peripheral blood cells. Use at 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Light Sensitivity: Protect from light; tandem dye is sensitive to photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be verified empirically.

Target Information

CD1d is a heavy chain associated with Beta-2 microglobulin on cortical thymocytes. Independent expression has been shown on human intestinal epithelial cells. CD1d is the sole group 2 member of the CD1 family of MHC-like glycoproteins. It mediates the presentation of lipid and glycolipid antigens to T cells. The CD1 family genes are clustered on chromosome 1 and differ in localization and ligand specificity. CD1d localizes to late endosomes and lysosomes via a tyrosine-based motif in its cytoplasmic tail. Diseases associated with CD1D include Mycobacterium malmoense infection and autoimmune disease.
