Target Information
Mitochondrial deoxyguanosine kinase (DGUOK) is required for the phosphorylation of several deoxyribonucleosides and certain purine deoxykribonucleoside analogs widely employed as antiviral and chemotherapeutic agents. These analogs are used in treatment of lymphoproliferative disorders and require intracellular phosphorylation to triphosphates for activity. In mammalian cells, phosphorylation of purine deoxyribonucleosides is mediated mainly by cytosolic deoxycytidine kinase (DCK) and mitochondrial deoxyguanosine kinase (DGUOK, also known as DGK). DGUOK is expressed ubiquitously, with highest levels in muscle, brain, liver, and lymphoid tissues. Defects in DGUOK cause mitochondrial DNA depletion syndrome (MDS), a heterogeneous group of disorders characterized by reduction in mitochondrial DNA copy number. MDS is inherited as an autosomal recessive trait and may affect single or multiple organs.
