Target Information
ERN1 (serine/threonine kinase) 또는 endoplasmic reticulum to nucleus signaling 1 단백질은 효모 Ire1 유전자의 인간 상동체로, 고유의 kinase 및 endoribonuclease 활성을 지니며 ER 기반 스트레스 신호에 관여합니다. ERN1은 포유류 세포에서 IRE1 단백질 분해를 조절하며, 태아 생존에 필수적인 세포 기능을 담당합니다. IRE1은 ER 내 비정상 단백질을 감지하여 XBP1 mRNA를 절단·활성화함으로써 unfolded protein response를 유도합니다. IRE1 결손 세포는 스트레스 조건에서 생존 불가하며, dominant negative IRE1 발현 시 ER 스트레스 하에서 세포사멸이 유도됩니다.
