Target Information
The Lipin family of nuclear proteins includes three members: Lipin-1, Lipin-2, and Lipin-3. Each contains a nuclear signal sequence, a conserved N-terminal (NLIP) domain, and a C-terminal (CLIP) domain.
LPIN1 (Lipin-1) is essential for normal adipose tissue development and metabolism. It selectively activates PGC-1A target pathways, such as fatty acid oxidation and mitochondrial oxidative phosphorylation, by inducing expression of the nuclear receptor PPARA.
LPIN1 also suppresses lipogenesis and circulating lipid levels. Overexpression promotes fat accumulation and insulin sensitivity, whereas deficiency may result in insulin resistance and lipodystrophy, characterized by loss of body fat, fatty liver, hypertriglyceridemia, and insulin resistance.
