Target Information

LATS2 is a serine/threonine protein kinase belonging to the LATS tumor suppressor family. It interacts with a negative regulator of p53 and functions in a positive feedback loop with p53 that responds to cytoskeleton damage, leading to centrosome and mitotic apparatus dysfunction. LATS2 plays a crucial role in maintaining mitotic fidelity and genomic integrity, as well as regulating organ size and tumor suppression by limiting proliferation and promoting apoptosis.

The LATS2 pathway involves a kinase cascade where STK3/MST2 and STK4/MST1, in complex with SAV1, phosphorylate and activate LATS1/2 with MOB1. Activated LATS1/2 then phosphorylate and inactivate YAP1 and WWTR1/TAZ oncoproteins. Phosphorylation of YAP1 by LATS2 prevents its nuclear translocation, thereby regulating genes involved in cell proliferation, apoptosis, and migration. LATS2 also negatively regulates the G1/S transition by downregulating cyclin E/CDK2 kinase activity.
