MLH1 is a DNA mismatch repair protein essential for maintaining genetic integrity. Defects in mismatch repair pathways can lead to microsatellite instability (MSI), which is associated with carcinogenesis. MLH1, together with PMSH and MSH2, participates in the recognition and repair of mismatched DNA during replication. Mutations in these genes are linked to hereditary nonpolyposis colon cancer (HNPCC).
