Target Information

NANOG (Nanog homeobox) is a divergent homeodomain protein that regulates pluripotency and differentiation in undifferentiated embryonic stem cells. NANOG mRNA is present in pluripotent mouse and human cell lines but absent in differentiated cells. Human NANOG protein shares 52% amino acid identity with the mouse protein and 85% identity in the homeodomain.

Human NANOG maps to gene locus 12p13.31, while the mouse NANOG maps to 6F2. Murine embryonic NANOG expression is detected in the inner cell mass of the blastocyst. Studies show high levels of NANOG expression in undifferentiated N-Tera embryonal carcinoma cell lines.

NANOG functions as a transcription regulator involved in embryonic stem (ES) cell proliferation and self-renewal. Its role in embryonic development suggests potential applications in generating stem cells for regenerative medicine and degenerative disease treatment.

Induced pluripotent stem (iPS) cells can be generated by expressing transcription factors such as NANOG, Sox2, Klf4, Lin28, and c-Myc. Overexpression of NANOG promotes S-phase entry and proliferation. Dysregulation of NANOG is associated with teratocarcinoma and germ cell/embryonal cancer.
