HFE2는 철 대사에 관여하는 단백질로, hepcidin 활성화 신호 경로의 구성 요소이거나 hepcidin 발현 조절자로 작용할 수 있습니다. 또한 hepcidin의 세포 수용체 역할을 할 가능성이 있습니다. 이 유전자에서는 서로 다른 isoform을 인코딩하는 대체 스플라이싱 전사체가 확인되었습니다.
HFE2 유전자의 결함은 hemochromatosis type 2A (juvenile hemochromatosis, JH)의 원인으로, 심각한 철 과부하로 인한 조기 발병 자가열성 질환이며, 30세 이전에 hypogonadotrophic hypogonadism, 간 섬유증 또는 간경변, 심근병증을 유발할 수 있습니다.
