Recommended positive control tissue: Parathyroid (not limited to this tissue type).

The antibody is intended for professional laboratory use in detecting the corresponding protein in formalin-fixed, paraffin-embedded tissue by manual qualitative IHC testing. It is used following primary diagnosis by conventional histopathology.

Background on PTH:
Parathyroid hormone (PTH) is produced in the parathyroid gland as an 84 amino acid single-chain polypeptide. It can be secreted as N-terminal or C-terminal fragments after intracellular degradation, such as in hypercalcemia.
Defects in the PTH gene cause familial isolated hypoparathyroidism (FIH), characterized by hypocalcemia and hyperphosphatemia due to inadequate secretion of parathyroid hormone. Symptoms include seizures, tetany, and cramps. FIH exists in both autosomal dominant and recessive forms.

Usage Notes:

  • Use with formalin-fixed, paraffin-embedded sections.
  • Pretreatment with heat-induced or enzymatic epitope retrieval recommended.
  • IHC staining visualizes antigens through sequential application of primary antibody, secondary antibody, enzyme complex, and chromogenic substrate.
  • Results are interpreted microscopically to aid in differential diagnosis.

Controls:

  • A positive tissue control must be run with every staining procedure.
  • Positive control tissues should include both positive and negative staining components.
  • External positive controls should be freshly fixed and processed similarly to patient samples.
  • Weak positive tissues are preferred for detecting minor reagent degradation.
  • Internal or external negative controls should demonstrate absence of specific staining.

Target Information

This gene encodes a member of the parathyroid protein family. The processed protein binds to the parathyroid hormone/parathyroid hormone-related peptide receptor and regulates calcium and phosphate levels.
Excess production leads to hyperparathyroidism and hypercalcemia; defective processing results in hypoparathyroidism and hypocalcemia.
