LanthaScreen™ TR-FRET LXR (liver X receptor) alpha Coactivator Assay 는 coactivator recruitment 작용자로 잠재적 LXR alpha ligand를 검사하는 민감도와 강건성이 높음 high-throughput 방법을 제공합니다. 이 kit는 혼합해 판독하는 균질한 분석 형식으로 terbium (Tb)-labeled anti-GST 항체, fluorescein-labeled coactivator peptide, glutathione-S-transferase (GST)로 태그된 LXR alpha ligand-binding domain (LBD)를 사용합니다.

LanthaScreen™ TR-FRET LXR alpha Coactivator Assay에는 fluorescein-coactivator peptide와 Tb-labeled anti-GST 항체 혼합물 첨가 후 LXR alpha-LBD를 ligand 검사 성분에 추가합니다. 실온에서 배양 후 520 nm/495 nm TR-FRET 비율을 계산하고 이를 성분의 용량반응곡선에서 EC50 결정에 이용할 수 있습니다. LXR alpha-coactivator peptide 상호작용 생물학에 기반을 둔 이 ligand EC50 는 수용체에 결합해 입체구조 변화에 영향을 미치고 coactivator peptide 모집에 필요한 ligand 양을 나타내는 복합 수치입니다(그림 1).

구성 및 보관:
