Background
The splicing pattern of TAL1/2 is cell-lineage dependent.
A chromosomal aberration involving TAL1 may cause certain T-cell acute lymphoblastic leukemias (T-ALL), particularly the translocation t(1;14)(p32;q11) with T-cell receptor alpha chain (TCRA) genes.
- Domain: The helix-loop-helix domain is necessary and sufficient for interaction with DRG1.
- Function: Implicated in the genesis of hematopoietic malignancies; may play an important role in hematopoietic differentiation and act as a positive regulator of erythroid differentiation.
- Post-translational Modifications (PTM):
- Phosphorylated on serine residues (notably Ser-122 under hypoxia).
- Ubiquitinated following hypoxia-dependent phosphorylation of Ser-122, leading to rapid degradation via the ubiquitin system (not observed in large vessel endothelial cells).
- Similarity: Contains one basic helix-loop-helix (bHLH) domain.
- Subunit: DNA binding requires dimerization with another bHLH protein. Forms heterodimers with TCF3 and binds to LMO2 and DRG1. Can assemble in a complex with LDB1 and LMO2 (TAL-1 complex: CBFA2T3, LDB1, TAL1, TCF3).
- Tissue Specificity: Leukemic stem cell.
