세포 내 위치 (Cellular Localization)

Cytoplasm and nucleus. Shuttles between the nucleus and the cytoplasm. Translocated into the nucleus upon tyrosine phosphorylation and dimerization in response to activated FGFR1–4 signaling. Constitutive nuclear presence is independent of tyrosine phosphorylation. Predominantly cytoplasmic without stimuli. Upon LIF stimulation, accumulates in the nucleus. The BART–ARL2 complex is important for nuclear translocation and retention of STAT3. Identified in a complex with LYN and PAG1.

Background

The protein encoded by this gene belongs to the STAT protein family. In response to cytokines and growth factors, STAT proteins are phosphorylated by receptor-associated kinases, then form homo- or heterodimers that translocate to the nucleus to act as transcription activators. STAT3 is activated by IFNs, EGF, IL5, IL6, HGF, LIF, and BMP2, mediating the expression of various genes involved in cell growth and apoptosis. Rac1 GTPase binds and regulates STAT3 activity, while PIAS3 acts as a specific inhibitor. Mutations in STAT3 are linked to infantile-onset multisystem autoimmune disease and hyper-IgE syndrome.
