Background
This gene belongs to the Tre-2/BUB2/CDC16 domain family. The encoded protein is a Rab-GTPase-activating protein containing two phosphotyrosine-binding domains (PTB1 and PTB2), a calmodulin-binding domain (CBD), a Rab-GTPase domain, and multiple AKT phosphomotifs.
It plays an essential role in glucose homeostasis by regulating insulin-dependent trafficking of GLUT4, which mediates glucose uptake into skeletal muscle and adipose tissue.
Reduced expression increases GLUT4 at the plasma membrane, indicating its role in intracellular GLUT4 retention under basal conditions. Upon insulin stimulation, the protein is phosphorylated, dissociates from GLUT4 vesicles, and promotes glucose transport.
