Product Specific Information

The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by thymocytes, mature T cells, and dendritic cells. CD4 binds to MHC class II and associates with protein tyrosine kinase p56lck through its cytoplasmic tail. Binding of GK1.5 is blocked by RM4-5.

Each product contains:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤0.2 µg/test (100 µL final volume). Optimize antibody titration for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample or per 100 µL cell suspension (10³–10⁸ cells).
• NovaFluor conjugates utilize Phiton™ technology for engineered fluorescent signatures minimizing spillover and spread.

Target Information

CD4 antigen is involved in recognition of MHC class II molecules and serves as a co-receptor for HIV. Primarily expressed in T helper cells, monocytes, macrophages, and dendritic cells. CD4 expression is found in thymus, lymph nodes, tonsils, spleen, and specific regions of brain and gut. It functions in early T-cell activation and may mediate neuronal damage in infectious or immune-mediated CNS diseases.
