Target Information

C12orf66 functions as part of the KICSTOR complex in the amino acid-sensing branch of the TORC1 signaling pathway.
It recruits the GATOR1 complex to lysosomal membranes, enabling its interaction with GATOR2 and RAG GTPases.
Acts upstream of RAG GTPases and negatively regulates mTORC1 signaling in the absence of amino acids.
Without the KICSTOR complex, mTORC1 remains constitutively localized to the lysosome and activated.
The KICSTOR complex may also participate in glucose-dependent regulation of mTORC1.
