Background
T cell receptors recognize foreign antigens processed as small peptides bound to MHC molecules on antigen-presenting cells (APC). Each T cell receptor is a dimer of one alpha and one beta chain or one delta and one gamma chain.
The TCR loci are rearranged and expressed sequentially (delta, gamma, beta, alpha). If delta and gamma chains are functional, the cell expresses them; otherwise, beta and alpha loci are rearranged.
Both alpha and delta loci include V (variable), J (joining), and C (constant) segments, while the delta locus also includes D (diversity) segments and is located within the alpha locus.
During T cell development, recombination joins D and J segments, followed by V-DJ joining for delta chains and V-J joining for alpha chains. C segments are added later at the RNA level.
This recombination and junctional diversity generate a wide range of antigen recognition.
Five variable segments can be used in either alpha or delta chains (TRAV/DV). Some V and J segments are pseudogenes incapable of encoding proteins.
