Target Information
SHP-1 (PTPN6)는 주로 조혈세포에서 발현되는 비수용체형 단백질 티로신 인산화효소입니다. 이 효소는 두 개의 SH2 도메인, 티로신 인산화효소 촉매 도메인 및 카복시 말단 조절 도메인으로 구성되어 있습니다. SHP-1은 여러 티로신 키나아제 경로를 음성 조절하기 위해 표적 단백질에서 인산기를 제거합니다. 조혈세포에서는 SHP-1의 아미노 말단 SH2 도메인이 인산화된 erythropoietin receptor(EpoR)에 결합하여 성장 신호를 억제합니다. 상피세포에서 SHP-1 과발현은 Ros receptor tyrosine kinase의 탈인산화를 유도하여 Ros 의존적 세포 증식 및 변형을 억제합니다. 또한, 골수세포에서 리간드 결합 후 SHP-1은 IL-3R 베타 체인과 결합하여 IL-3 유도 인산화 및 세포 증식을 하향 조절합니다.
