Target Information
ERN1 (serine/threonine kinase), also known as endoplasmic reticulum to nucleus signaling 1, is a human homologue of the yeast Ire1 gene product. It possesses intrinsic kinase and endoribonuclease activities, playing a crucial role in ER-based stress signaling. ERN1 regulates IRE1 proteolysis in mammalian cells and is essential for fetal viability through its function in extraembryonic cells.
IRE1 acts as a sensor for unfolded proteins in the ER, initiating the unfolded protein response by splicing XBP1 mRNA into an active transcriptional activator. While null mutants are viable, IRE1 is critical under stress conditions that cause unfolded protein accumulation in the ER. Depletion of IRE1p does not affect transfected cells under normal conditions but leads to apoptosis under ER stress.
