Target Information
Human MTM1 is a 603 amino acid protein mutated in myotubular myopathy. The related protein hMTMR2 is mutated in a recessive form of Charcot-Marie-Tooth neuropathy.
Myotubularin functions as a lipid phosphatase acting on phosphatidylinositol 3-monophosphate, regulating the PI3-kinase pathway and membrane trafficking. Wild-type myotubularin can dephosphorylate PI 3-P and PI 4-P in vitro, thereby decreasing PI 3-P levels by down-regulating PI3-K activity and promoting PI 3-P degradation.
