Target Information
Tumor necrosis factor (TNF) receptor-associated factors (TRAFs) are adaptor proteins that link the TNF receptor superfamily to signaling pathways, regulating cell death and stress responses. TRAF proteins share a conserved region enabling receptor and TRAF binding, activating diverse signal cascades.
TRAF2 and TRAF3 bind directly to CD40, a receptor critical for NF-κB activation in B lymphocytes. TRAF2, along with TRAF6, is required for CD40 signaling in non-hematopoietic cells. TRAF2 also interacts with the lymphotoxin-beta receptor (LTβR) alongside TRAF3 and apoptosis inhibitors cIAP1 and Smac.
