Beta-catenin is an adherens junction (AJ) protein essential for cell-cell adhesion and epithelial layer maintenance. It interacts with E-cadherin and alpha-catenin, linking them to the actin cytoskeleton. Beta-catenin also associates with complexes containing APC protein and microtubules, and participates in signaling pathways involving tyrosine kinases, phosphatases, and Wnt/Wingless. These interactions regulate morphogenesis and cell proliferation.
Beta-catenin is expressed in various hair follicle cell types and epithelial tissues. It has been implicated in several cancers, including pilomatrixoma, medulloblastoma, colorectal, ovarian, and other solid tumors. In the nucleus, beta-catenin co-activates Lef/Tcf transcription factors, promoting expression of genes such as cyclin D and c-myc that drive cell proliferation.
