Target Information

SHANK3 is a major scaffold postsynaptic density protein that interacts with multiple proteins and complexes to orchestrate dendritic spine and synapse formation, maturation, and maintenance. It interconnects postsynaptic receptors including NMDA-type and metabotropic glutamate receptors via complexes with GKAP/PSD-95 and HOMER, and links to the actin-based cytoskeleton. SHANK3 plays a role in dendritic spine structural organization through interactions with Arp2/3 and WAVE1 complex, promoting F-actin clusters. It regulates neuron growth cone motility and NMDA receptor signaling, modulates GRIA1 exocytosis and GRM5/MGLUR5 expression, controlling synaptic transmission and plasticity. SHANK3 may be required at early synapse formation stages and inhibited by IGF1 to promote maturation. (Uniprot)
