Target Information
FOXO1 (FKHR, Forkhead Box O1)는 전사인자(forkhead homeotic gene family)의 하위군에 속하는 단백질로, 핵 수용체 활성 조절에 관여합니다. FOXO1은 PAX3와의 상호작용을 통해 alveolar rhabdomyosarcoma와 관련이 있으며, 항종양 활성, 자가포식, 근육 성장 및 분화 과정과 연관되어 있습니다.
FOXO1은 인슐린 신호전달의 주요 표적 단백질로, 산화 스트레스에 대한 대사 항상성을 조절합니다. 또한 FOXO1은 IRE (Insulin Response Element) 및 DBE (Daf-16 Binding Element)에 결합하여 전사 조절에 관여합니다. 골 형성과 적색산화 균형 조절, 골격계의 내분비 기능, 포도당 대사 조절, 그리고 RUNX2 억제를 통해 osteocalcin/BGLAP 발현을 제어합니다. 간세포에서는 PPARGC1A와 함께 gluconeogenesis 관련 유전자(IGFBP1, G6PC, PPCK1)를 활성화합니다. FOXO1은 CDK1, PKB/AKT1, SKT4/MST1 하류에서 세포사 조절에도 관여합니다.
