Target Information
Estrogen Receptors (ER) belong to the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, upon estrogen binding, undergoes conformational change enabling dimerization and DNA binding to estrogen response elements (ERE). ER regulates gene transcription positively or negatively through coactivator or corepressor recruitment.
Two forms exist: alpha (ESR1) and beta (ESR2), encoded by separate genes. Alternative RNA splicing yields at least four alpha isoforms (66, 53, 47, and 35 kDa).
ERs are widely expressed across tissues, essential for sexual development and reproduction, and also involved in bone physiology. Dysregulation contributes to diseases such as breast cancer, endometrial cancer, and osteoporosis.
