The Human B7-H6 Simplex ProcartaPlex™ Kit measures B7-H6 protein and is designed to be combinable with other Simplex kits, allowing creation of custom multiplex panels utilizing Luminex xMAP technology for protein detection and quantitation. When combining multiple Simplex kits (i.e., not using a pre-configured Multiplex Panel), only one buffer kit (sold separately) is needed per assay plate regardless of plex size.

About ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays are based on sandwich ELISA principles, using two highly specific antibodies binding to different epitopes of a protein to quantitate all targets simultaneously using a Luminex instrument. These multiplex assays require as little as 25 μL of plasma or serum, or 50 μL of cell culture supernatant, and results can be obtained within four hours.

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for simultaneous detection and quantitation of up to 80 protein targets in a single 25–50 μL sample from plasma, serum, cell culture supernatants, and other bodily fluids.

The Luminex beads are internally dyed with precise proportions of red and infrared fluorophores, creating spectrally unique signatures identifiable by Luminex xMAP detection systems (e.g., Luminex 200, FLEXMAP 3D, MAGPIX). Similar to a sandwich ELISA, the assay uses matched antibody pairs to identify the target protein. Each bead is labeled with antibodies specific to a single protein, and bound proteins are detected with biotinylated antibodies and streptavidin–R-phycoerythrin (RPE).

The main difference from ELISA is that the capture antibody is conjugated to a bead rather than adsorbed to a microplate well, allowing free-floating reagents in solution. The Luminex 200 instrument uses two lasers — one to distinguish bead signatures and another to quantify RPE fluorescence proportional to protein concentration. ProcartaPlex multiplex assays can profile more targets using less sample and time compared to traditional ELISA.
