Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein belonging to the immunoglobulin family of receptors. It plays an integral role in signal transduction and T cell differentiation and activation. CD8 is predominantly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with the T cell receptor (TCR) for MHC-I molecules. In contrast, CD4 acts as a co-receptor for MHC-II molecules.

CD8α is essential for binding to MHC-I and is also expressed as homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells. Engagement of MHC-I/peptide complexes by antigen-presenting cells (APCs) recruits lymphocyte-specific protein tyrosine kinase (Lck), leading to cytokine production, motility, and activation of cytotoxic T lymphocytes (CTLs). Activated CTLs are critical for pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
