Target Information

The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. It has three major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain. AR acts as a ligand-activated transcription factor that binds active testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, the receptor translocates into the nucleus, dimerizes, and activates transcription of androgen-responsive genes.

AR signaling is essential for the development and function of male reproductive organs such as the prostate and epididymis, and also functions in nonreproductive tissues including muscle, hair follicles, and brain. The androgen receptor is a phosphoprotein that regulates mitogen-activated protein kinase (MAP kinase) pathways. Dysregulation of AR signaling has been linked to diseases including prostate cancer, Kennedy’s disease, and male infertility. Mutations in the AR gene are associated with complete androgen insensitivity (CAIS).
