Target Information
Xeroderma pigmentosum type G (XPG) is a human genetic disease exhibiting extreme sensitivity to sunlight.
The XPG protein, a member of the flap endonuclease 1 (FEN-1) structure-specific DNA repair endonuclease family, is essential for DNA repair of solar UV-induced DNA damages.
- XPG nuclease performs the 3′ incision during nucleotide excision repair of DNA.
- The enzyme cleaves DNA bubble structures near the junction of unpaired and duplex regions.
- A 29-amino acid region (residues 981–1009) contains PCNA binding activity.
- Arginine 992 (Arg992) is critical for PCNA binding.
- Replication Protein A (RPA) binds directly to XPA and XPG, both involved in excision repair.
