Ion Torrent™ Oncomine™ Tumor Mutation Load Assay (Manual Library Preparation)

The Oncomine Tumor Mutation Load Assay is a targeted next-generation sequencing (NGS) assay that provides an assessment of tumor mutation load and mutation signatures in a simple workflow. It enables library construction from two pools of multiplex PCR primers using only 10 ng of DNA per primer pool from formalin-fixed paraffin-embedded (FFPE) tumor samples — without the need for a matched normal sample.

This assay helps identify and select samples most likely to respond in cancer immunotherapy research.

The resulting libraries are compatible with automated template preparation and chip loading on the Ion Chef™ System.

From Sample to Answer

The assay integrates with optimized informatics and visualization software for a complete sample-to-answer workflow.
Run metrics can be analyzed using the Torrent Suite™ plugin.
The Ion Reporter™ workflow uses a custom variant calling and germline filtering algorithm to estimate somatic variants accurately.
Reports include normalized mutation load (mutations/MB) and mutation signatures such as UV damage, tobacco smoke damage, de-amination, and substitution patterns.
