The Thermo Scientific™ GlycoLink™ Immobilization Kit provides affinity columns and reagents designed to immobilize glycoproteins through oxidized sugar groups for use in affinity purification or protein–protein interaction studies.

Description
The kit uses hydrazide-activated resin and a coupling catalyst to prepare affinity columns with polyclonal or monoclonal antibodies containing periodate-oxidizable carbohydrate groups. The resulting immobilized antibodies maintain unobstructed antigen-binding sites, enabling optimal purification performance.
Prepared columns can be regenerated and reused at least five times without significant loss in binding capacity.

Principle of Operation
Glycoproteins, such as antibodies, contain carbohydrate moieties with cis-diol groups that can be oxidized by sodium meta-periodate to form aldehydes. These aldehydes react with hydrazide groups on the resin to form stable covalent bonds. The GlycoLink Coupling Catalyst accelerates this reaction, enhancing coupling efficiency and reducing incubation time.
Typical coupling reactions occur within 2–4 hours in a non-amine buffer, achieving >85% efficiency and yielding 1–10 mg of immobilized protein per mL of UltraLink™ Hydrazide Resin.
